![]() ![]() ![]() Sequence consensus analysis of BAP1-bound sites showed enrichment of motifs specific for YY1, NRF1 and Ets transcription factors, which have been shown to interact with BAP1 in other cell types. BAP1-bound genomic loci showed narrow distributions of insertions and were near transcription start sites, consistent with recruitment of BAP1 to these sites by specific DNA-binding proteins. We found a strong correlation between multiple calling-card transposon insertions targeted by BAP1-PBase and BAP1-responsive expression of adjacent genes. Sets of significant genes were analyzed for common pathways, transcription factor binding sites, and ability to identify molecular tumor classes. We also examined RNA expression in the same OCM-1A UM cells after BAP1 depletion to identify BAP1 binding sites associated with BAP1-responsive genes. The insertion of transposons near BAP1 binding sites in UM cells were identified by genomic sequencing. The transposase piggyBac (PBase) was fused to BAP1 and expressed in OCM-1A UM cells. This system was developed to identify the genomic loci visited by transcription factors that bind directly to DNA our study is the first use of the system with a chromatin-remodeling factor that binds to histones but does not interact directly with DNA. We have used the “Calling Card System” of transposase-directed transposon insertion mapping to identify the genomic targets of BAP1 in uveal melanoma (UM). Keep it simple, show only your best work, show only what people want to buy, and you will be a success.BAP1 is a histone deubiquitinase that acts as a tumor and metastasis suppressor associated with disease progression in human cancer. However, those get lost amongst the photos of your sister, a light pole, and other non-sellable photos. ![]() Even if people did scratch the "f" word onto a tree in your picture, you don't want that word on your site.Ī photo of someone else's work of art (for example, the foot statue) shouldn't be on your site.įrom looking at your site, I can see that you are very creative and try new techniques, which I find fantastic. If you want to display photos of your family, don't indicate their relationship to you on the site.ĭon't use profanity on your site. This makes people think that you are just starting out and can't get clients. NEVER NEVER use the word "thing" on a professional site: "a flower bouquet type thing in downtown chicago"Ī lot of your photos show family members. If it's "just" some beads, why do I want to look at it? For example, you have a description saying "just some beads". You should also never downplay your work. Never, never put something below average on your business site. Why location? How will they know if they can hire you if it doesn't say where you are? Then, they should see your VERY BEST work. When someone logs onto your site, they should see your business name, contact information (NOT myspace), and location. I suggest just placing a title below the picture, not a description of what you did, unless it's necessary (like for the photo retouching one).Īs others have said, myspace accounts do not say "professional." Get rid of that link, or place it at the bottom of your page. Almost all of your photo descriptions are this way. the more of her, the merrier" below a photograph makes me think this is more of a myspace type of photo gallery. Next, placing statements such as "kind of a random shot of my grandma from photo one. Why do you have your color contact sheet there? And, EEK, it shows that you get things printed at Walgreens!! Not something you want to advertise, believe me! Nothing says amateur like bad grammar and punctuation. You need to use proper English, including capitalizing the first word of a sentence and ending with a period. If that is the case, then you still have some work to do. Since you're posting in the Business section of this forum, I assume you want your website to be used to make you money. Google dropped the ball on this application, in my opinion. For some reason, they make it almost impossible to place two pictures side by side, and if you do manage to get them that way, when you save your page, it moves things around. I've tried it and it was such a pain in the rear that I hardly even use it anymore. Google falls waaaaaaaay short when it comes to displaying pictures on googlepages. ![]()
